چکیده

The sensitising potential of chemicals is usually identified on the basis of animal studies. In addition to increasing public concern regarding the use of animal testing for the toxicological evaluation of new chemicals, the adoption of the 7th Amendment to the Cosmetics Directive in Europe will result in animal testing of ingredients used in cosmetics being subjected to severe restrictions by 2009 which will be tightened by 2013. The development of in vitro models for predicting the sensitising potential of new chemicals is therefore an obvious need. In vitro sensitisation tests will have to reflect the complex interactions of a chemical with the different compartments of the immune system. COLIPA STTF is therefore supporting a range of research projects reflecting our current understanding of the molecular and cellular events occurring during the sensitisation process from aspects of chemistry, peptide binding and skin metabolism to intracellular signalling pathways induced by allergens (see fig. 1).

Comparison of Functional Competence of Umbilical Cord and Adult Peripheral Blood Dendritic Cells in Allogenic Mixed Leukocyte Reaction

Background: Dendritic cells (DCs) are the most potent stimulators of primary T cell responses and play a key role in immune reactions after stem cell transplantation. Very little is known about the cord blood (CB) dendritic cells and their potential involvement in the low incidence and lower severity of acute graft-versus-host disease after CB transplantation.   Objectives: The aim of this stud...

A Shot at Dendritic Cell-Based Vaccine Strategy against HIV-1

Introduction: Despite considerable efforts to control AIDS pandemic, it is still one of the significant infectious concerns worldwide. The advance in medical research has led to the development of highly active antiretroviral therapy with a considerable effect to suppress the disease. However, an effective vaccine capable of eradication the HIV pandemic is not available yet. Failure to develop ...

The Effect of Interferon Beta in HLA-G Expression on Monocyte in Diabetes Type1

Abstract Background and objectives: Dendritic Cells are the most important of antigen presenting cells with an effective role in immune tolerance. This study, aims to clarify the role of IFN- β in induction on dendritic cells derived by monocyte in diabetes type1 to evaluate the T cells response to beta cell specific antigenic molecule. Material and Methods: In this research, peripheral bl...

Comparison of Several Maturation Inducing Factors in Dendritic Cell Differentiation

Background: Dendritic cells (DCs) are professional antigen presenting cells that have an important role in the initiation of immune response. The use of maturation factors in dendritic cell differentiation provides a promising approach in immunotherapy. Objective: In this study, we compared tumor necrosis factor-α, polyribocytidylic acid, lipopolysacharide and CpG oligonucleotides in inducing d...

The Effects of Candida Albicans Cell Wall Protein Fraction on Dendritic Cell Maturation

Back ground: Candida albicans is a member of the normal human microflora. C. albicans cell wall is composed of several protein and carbohydrate components which have been shown to play a crucial role in C. albicans interaction with the host immune system. Major components of C. albican cell wall are carbohydrates such as mannans, β glucans and chitins, and proteins that partially modulate the h...

Highly Efficient Transfection of Dendritic Cells Derived from Esophageal Squamous Cell Carcinoma Patient: Optimization with Green Fluorescent Protein and Validation with Tumor RNA as a Tool for Immuno-genetherapy

This study was conducted to optimize a highly efficient mRNA transfection into dendritic cells (DC) derived from esophageal squamous cell carcinoma (ESCC) patients. Applying an electroporation technique, in vitro synthesized Green Fluorescent Protein (GFP) mRNA was transfected as an indicator into the DCs derived from a healthy donor. Flow cytometry revealed 84.9% transfection efficiency for DC...
